Cardiovascular Genetics: A News Round-Up Selected News Items Summarized by Wolfgang Lieb, MD; Ramachandran S. Vasan, MD Mutations in Sarcomeric Protein Genes Account for a Substantial Proportion of Idiopathic Cardiac Hypertrophy in Children

چکیده
Principal Findings The authors identified mutations in almost half of children without a positive family history of cardiomyopathy (25 of 51 affected children) and in two thirds (21 of 33) of affected children with familial cardiomyopathy. Mutations in MYH7 (encoding -myosin heavy chain) and MYBPC3 (encoding myosin-binding protein C) were the most frequent variants identified in the children. In the subgroup with sporadic (nonfamilial) cardiac hypertrophy, boys were more likely than girls to harbor mutations.

How Was the Hypothesis Tested? The authors performed a genome-wide linkage analysis and subsequent fine mapping in a family with 11 affected members to identify a chromosomal locus segregating with the condition. Then, candidate genes in the critical region were sequenced and biochemical analyses as well as animal experiments were performed to assess the biological consequences of the detected ...
متن کاملMolecular Modelling and Evaluation of Hidden Information in ABCB11 Gene Mutations
Background: Cholestatic disorders are divided in the extra and intra-hepatic that created due to the severe liver diseases. ABCB11 encodes the bile salt export pump and this gene is mutated in several forms of intrahepatic cholestasis. So far, some molecular features of this gene was studies. Objective: Using a developed web server, we identified high number of rare codons in this gene, and four...
متن کاملIdentification of a new missense mutation in MyBP-C associated with hypertrophic cardiomyopathy.
Hypertrophic cardiomyopathy is a primary cardiac disease, characterised by idiopathic myocardial hypertrophy, and is caused by defects in sarcomeric protein encoding genes. One of these genes is cardiac myosin binding protein C (MyBP-C), in which a number of splice site and duplication mutations causing HCM have been described. During mutation screening of a South African HCM population by PCR-...
متن کاملThe Importance of Electrocardiography in Pediatric Patients with Pulmonary Arterial Hypertension in Follow- up
Background: Right Ventricular (RV) hypertrophy is an adaptive response to chronic RV pressure overload in patients with pulmonary hypertension. We investigated the relationships between RV hypertrophy indicators, including electrocardiography, the percentage oxygen saturation (SaO2%), body mass index (BMI), and blood uric acid levels in patients with...
